About the project of Domenico

The overall goal of my my research work is the production of nanoparticles (called Nano-Ghosts, Ngs) from mesenchymal stem cells (MSCs) for targeting cartilage tissue regeneration. We aim to entrap different therapeutics (e.g growth factors, antibiotics, corticosteroids, etc.) in the NGs, and study their targeting and therapeutic efficacy in vitro and in vivo. Different physical and chemical characterization techniques will be used to investigate the properties of the NGs and to analyze the shape and the size of the particles needed as well as the entrapment and the release of drugs with different chemical properties. In vitro and in vivo tests will be performed to better understand particles behavior interaction with cells, release of the therapeutic from the NGs and their biological activity. In addition we will address other methods to release the drug when entrapping the NGs in a bioplatformed to be used for cartilage repair (use of soft matrix or gel) as well as indirect encapsulation of the drug using mediators.
